[RCI 12 June 1995] Data for the root-tissue bags used to generate a
transport loss and oven- to air-dried conversion factors, were
appended to the data section on 19 June 1995. Data for 7 bags
harvested in June 1994 were appended on 19 June 1995.[RCI 19 June 1995]
The incorrect processing program had been run on the 7 bags harvested
in June 1994 and processed on 19 June 1995, so that the weight loss
and percent weight loss values had not been calculated. These data
were reprocessed and the weight loss and percent weight loss values
were added to the appropriate locations in the data section on 23 June

TITLE. Litter and root decomposition in alpine tundra.
ABSTRACT. Plant tissues, foliage, and roots were collected from
Niwot Ridge tundra species and composited into separate 2-gm samples.
Foliage was placed in 10x20 cm litter bags with a 2mm^2 mesh to
allow entry of invertebrates. Root tissues were placed in 10x20
cm^2 polyester fabric bags to prevent loss of fine root particulates.
The foliage litter bags were placed on the tundra surface and
secured by 10-penny nails. Root bags were buried in slits 15 cm
deep. Each of the 18 1-m^2 research plots, consisted of 2 rows of
5 surface litterbags each. The upper row (with respect to slope)
served as the control. The lower row was treated with ammonium
nitrate at a rate of 20 g per m^2. Nitrogen will be applied at
3 monthly intervals during each of the growing seasons for the duration
of the study. Litter bags (1 surface and 1 buried from each
control and treatment) were collected from each plot on 18 September
1993. Additional collections are to be made in spring and fall of
1994 and spring of 1995. The samples are to be analyzed for changes
in total mass, N concentration, and variation in C fractions.

COMMENTS. Plant tissue samples were dried and ground to a fine powder
and analyzed via modified ADF technique (Effland 1977) for cellulose,
non-polar carbon and lignin.
Data were manually entered using EasyEntry forms and processed on

Calculated numbers in this file may differ slightly from published
numbers due to differences in rounding techniques.
Bag numbers 1 through 179 represented the mesh bags and bag
numbers > 179 represented tissue polyester bags. Bags 180 through
210 represented foliage surface bags and were part of an experiment
to test the effects of bag material. Bag numbers >210 represented
root tissue bags that were buried at a depth of 15 cm.
Nitrogen treatments are high (H), low (L), or "mixed" (X). The high
treatment consisted of 20 g N/m^2/month applied 3 times during the
growing season (one application each in July, August, and September)
of each year. This treatment was delivered as an aqueous solution of
ammonium nitrate. The low treatment represented the control, i.e.,
ambient, naturally-occuring levels of nitrogen. The "mixed" treatment
also represented a control with respect to nitrogen. However, this
treatment was distinguished from the L treatment by smaller (<0.1 mm)
mesh-size bags.
Moisture "treatments" are dry (D), mesic (M), or wet (W).
Depth treatments are surface (S) or belowground (B).
Data for which the field placement data and the harvest date were
the same (1 July 1993), were used to characterize the litter weight
loss effected by transport of litter bags to the field and then
back to the lab. Percent weight loss was calculated for each of
these bags and a mean percent weight loss was calculated from these
values (excluding data for bags 146, 182, and 186 which were deemed
to be outliers) for both the foliage litter and the root tissue
bags. Surprisingly, mean percent weight loss was higher for the
polyester bags (4.21% vs 2.67%) and it was discovered that the initial
and final samples were weighed air-dried and oven-dried, respectively.
A more accurate mean percent weight loss can be calculated by
establishment of the relationship between air-dried and oven-dried
weights for this type of biomass material. At any rate, the mean
percent transport weight loss was then applied to generate a
corrected initial dry litter weight:

to get expected weight loss due to transport, and
COL12 = (COL6 - staple weight - COL3) - expected weight loss
where staple weights were 0.06 and 0.09 g for litter mesh and polyester
bags, respectively.
This corrected weight was then used to calculate decomposition for
subsequent harvests.
Litter weight loss (COL16) was calculated as
COL12 - COL14.
Percent weight loss (COL17) was calculated as
(COL16/(COL12))*100.
Non-sample weight change (COL18) was caculated as
COL15 - COL14 - COL3.
Note that any additional staple weights were included in the
final total weights during data entry, with the weight of 1
staple=0.03 g.
Also note that calculated columns COL12, COL16, and COL17 are
meaningless where field placement date and harvest date are the same
(i.e., 07/01/93) since the correction factor for weight loss during
transport has, in effect, been applied twice.
Dave Bryant established plot locations on 16 June 1994 (TD1,TM1,TW1) and
8 July 1994 (remaining plots). A Silva Ranger compass was used with E
declination set at 11.5 degrees; the accuracy of the bearings is +/-
3 degrees. All distances were measured across the ground surface (not at
a fixed horizontal level); such distances are accurate to +/- 2%. The plot
code, bearing (degrees) from indicated reference point to plot, and
distance (meters) from indicated reference point to plot follow:

It is believed that the plot code currently listed as 26W in the data
section is actually SW3, but this was unable to be verified for certain
until the summer of 1995.

Carbon fraction extraction for the plant litter samples was accomplished
using an apparatus identical to that described in Van Soest and Goering
(1970). The technique was a modified Van Soest from Effland (1977). Prior
to analysis of litter samples, preliminary analyses were performed using
standards provided by John Aber's Laboratory at the University of New
Hampshire's Center for Complex Systems Research. The standards consisted of
oven-dried, ground foliage samples of pre-senescent sugar maple (Acer
saccharum), black locust (Robinia pseudoacacia), and red oak (Quercus rubra).
Once the method was determined to faithfully reproduce the known carbon
concentrations for these species, an internal lab standard (Plantago spp.)
was calibrated against them for future use. The process is a proximate
gravimetric analysis where the material is subjected to successive
digestions in specific solvents to remove the compound of interest.
Following each digestion, the sample is weighed. The loss of weight from
each step is attributed to the constituents removed.
For non-polar constituents, the plant sample was placed in a 50-ml low form
Gooch crucible that had been ashed, and pre-weighed. The total weight of the
sample and crucible was recorded. The crucible was placed in a 250-ml
Berzelius beaker and 150 ml of dichloromethane were added. The beaker was
placed on a hot plate and fitted with a cold water reflux condenser.
Samples were boiled for 3 hours, removed, and allowed to stand overnight
to allow evaporation of the solvent (covered with a fine mesh screen to
prevent contamination). The samples were oven-dried at 60 degrees Celsius
and weighed. This step removes waxes, phenols, chlorophyll, and other
non-polar substances. Many substances in plant tissues (amino acids and
sugars) are known to be soluble in both polar and non-polar solvents. The
vessel used for boiling required a larger volume of solvent to contact the
sample. It was believed that this larger volume extracted the non-polar
substances more efficiently and that polar substances were being extracted
as well. For this reason, the boiling time was reduced from 5 hours to 3
hours. The reduction in boiling time reproduced the known values for the
calibration standards (maple, locust, and oak).
For polar constituents, it is necessary that the water-soluble C fractions
(sugars, amino acids, and proteins) be removed. Following the weighing of
the sample from the previous digestion, the crucible and sample were placed
in a 250-ml beaker and refluxed in 200 ml of de-ionized water for 5 hours.
The samples were removed and vacuum filtered, washed with de-ionized water
for 15 minutes, and oven-dried. Dry weights of sample and crucible are
recorded.
For acid-soluble constituents, it is necessary that the cellulose and
hemicellulose be removed, leaving acid-insoluble residue (lignin). A 500-g
subsample from the previous digestion was placed in a dried 30-ml test tube,
and 7.5 ml of concentrated sulfuric acid were added. The test tube was
placed in a 30-degree Celsius water bath for 30 minutes and stirred with a
glass rod. The sample was washed from the test tube into a 500-ml beaker,
adding sufficient de-ionized water to bring the volume to 210 ml. The
addition of water produced 72% sulfuric acid. The sample was then refluxed
for 5.5 hours. Additional de-ionized water was added following digestion to
cool the sample and arrest the reaction. The sample was vacuum filtered
through a pre-ashed and weighed crucible and washed with de-ionized water
for 15 min. The sample and crucible were oven-dried at 60 degrees Celsius
and weighed.
Subsamples were removed from the 25 September 1994 harvest litter bags in
order to ascertain the relative proportion of fungal to bacterial biomass
on the litter. These subsamples were removed immediately upon retreival and
prior to drying. They were shipped overnight express to Elaine Ingham's
Laboratory at Oregon State University. Because there was limited funding
available for this analysis, control samples of surface and buried litter
were sent from all 3 landscape placements. Treatment samples for surface
litter bags for dry and wet communites only were submitted for analysis.
Because the subsamples for microbial analysis were removed prior to drying,
there was no final measured dry litter weight (COL14) for these samples.
Instead a final dry litter weight (COL23) was derived from the ratio of
dry to wet litter weights (using the weights from the portion of the sample
not used for microbial analysis, i.e., COL20 and COL21) and the sum of the
wet weights of the subsample used for microbial analysis and the portion
of the sample not used for microbial analysis (i.e., COL19+COL20).
Similarly, a dry litter weight of the subsample used for microbial
analysis (i.e., COL22) was derived.
David Bryant supplied preliminary nitrogen determinations for litter
samples in mid-January 1995. Redeterminations using a C/N analyzer were
expected to replace these data, listed as litter bag number and percent
nitrogen:

In late March 1995, Bryant noticed that for buried root tissue samples
(i.e., bag numbers >210) the values in columns 5 and 6 were not
actually measured and so should have appeared as being missing.
Previously those values had been derived in order to calculate the
value in column 12, but Bryant developed a conversion factor
(-15.3% of the initial root tissue weight, i.e., column 4) to account
for transport/handling losses, as well as the fact that the initial
weights were not oven-dry weights. The root-tissue bags from which
this conversion factor was developed are indicated by a field placement
date of 07/??/93 because the bags were not actually buried in (i.e., were
only transported to and from) the field. The data were corrected accordingly.
In addition, the labels for columns 4, 12, 14, 16-17, and 19-23 in the
header section were expanded to clarify the distinction between litter
and root tissue samples.
