TITLE. Luxury uptake and storage of nitrogen in the rhizomatous alpine
herb, Bistorta bistortoides. 

ABSTRACT. The patterns of luxury consumption and storage of nitrogen were 
studied in the rhizomatous alpine herb, Bistorta bistortoides (Pursh.). 
Emphasis was placed on understanding the mechanisms, carbon costs, and 
possible benefits to growth of assimilating and storing excess nitrogen. 
Nitrogen pools in the rhizome and shoot were measured in fertilized and 
non-fertilized plants over two growing seasons (the 3rd and 4th growing 
seasons after initiation of fertilization). Fertilized plants stored more 
N in the rhizome throughout both seasons and exhibited greater mobilization 
of N reserves to support growth, compared to non-fertilized plants. Flower, 
foliar, and preformed bud N concentrations and biomass production rate did 
not increase in response to fertilization. The increased storage of N 
without a concomitant influence on biomass production demonstrates that 
luxury consumption occurs in this species. Fertilized plants relied more 
heavily on stored N to drive seasonal growth, with translocation from 
reserves accounting for 56% and 100% of the aboveground N requirement in 
non-fertilized and fertilized plants, respectively. In fertilized plants, 
the complete reliance on stored N indicates that the increase in 
utilization of N from reserves is accompanied by a decrease in utilization 
of N from current-season uptake. Total plant N content in fertilized 
plants did not change throughout the entire growing season, demonstrating 
the absence of net N uptake from the soil. Thus, the uptake and storage of 
excess N was accompanied by negative feedback to further soil N uptake. 
Such feedback would constrain the continued accumulation of excess N and 
eliminate any long term benefits of luxury N uptake in this species.
Increased N storage in fertilized plants was accommodated by an increase 
in the amino acids arginine, the non-protein amino acid, 
delta-acetyl-ornithine, glutamine, and glutamate. It was found that the 
entire length of the rhizome, including tissues that are several decades 
old, and both the pith and the cortex are active in N storage. Luxury 
uptake was accommodated entirely through filling of existing rhizome cells; 
no increased cell proliferation was found. Sucrose concentration was 
significantly lower in the recently produced rhizome tissue of fertilized 
plants, reflecting the increased, direct carbon costs of storing the 
additional N.
There do not appear to be growth benefits associated with luxury N uptake 
in this species. In response to luxury uptake a switch occurs, such that 
growth is driven entirely by stored, excess N, rather than a combination 
of stored N and N assimilated during the current season. By relying 
entirely on stored N, the cost of root construction and nutrient 
assimilation should be reduced. However, this apparent benefit is offset 
by (1) inherent developmental constraints to the rate of growth in this 
species which prevents the utilization of excess N to drive increased 
growth, (2) negative feedback between luxury uptake and future N 
assimilation, and (3) substantial direct carbon costs of storing the 
excess N. These costs and constraints appear to balance any benefits to 
luxury uptake, resulting in no net response of plant growth.
Delta-acetyl-ornithine was found to vary seasonally and in response to
nitrogen availability in the rhizomes of the alpine herb, Bistorta 
bistortoides (Pursh). Delta-acetyl-ornithine accounts for 38% of the free 
amino acid pool and up to 12% of the total nitrogen in rhizomes. It is 
concluded that this compound plays an important role in nitrogen storage 
in this species.

The site for this study was within a moist meadow community (as defined 
by May and Webber, 1982) dominated by the forb Acomastylis rossii and the 
graminoid Deschampsia caespitosa, and sub-dominated by B. bistortoides. 
The meadow lies on the western side of the saddle (the "Saddle") formed 
by two knolls on Niwot Ridge in the Front Range of the Colorado Rocky 
Mountains.
Observations were made on plants in N fertilized and control (non-
fertilized) plots. The fertilization treatment was applied over the 
two-year period of 1991-1992 as described previously (Bowman et al. 1995). 
Five replicate 1 m2 plots were fertilized with biweekly additions of 2 g 
NH4NO3 dissolved in 5 L of deionized water. The meadow surrounding the 
plots served as a non-fertilized, unwatered control. Bowman et al. (1995) 
found no effect of water alone on biomass or leaf gas exchange in Bistorta 
bistortoides in these plots. The current study was conducted during 1993 
and 1994, 1-2 years after fertilization had ceased.
In 1993, the first year after cessation of fertilization, plants from 
fertilized and non-fertilized plots were collected at the time of full 
bloom at mid-season (27 July) and at the time of leaf senescence at the 
end of the season (16 September). In 1994, fertilized and control plants 
were collected just after snowmelt when leaves were still unexpanded 
(25 May-1 June), at mid-season (6 July), when plants were in full bloom, 
and at the end of the season (3 September). Plants were also collected on 
18 August, when leaves were partially senesced, for the comparison of N 
concentrations in cortex and pith of rhizomes. Non-fertilized control 
plants were collected on two additional dates (22 June, 26 July) during 
the season. On each sampling date, 4-8 plants from both treatments were 
completely excavated from the soil. The rhizome and fine roots of B. 
bistortoides are rather shallow, making it easy to harvest the entire 
plant.
Within 6 hours of collection, plants were washed in distilled water, fine 
roots and previous years' dead tissue were removed, aboveground portions 
were separated from the rhizome and divided into leaves and the 
inflorescence (including the stalk and attached leaves) for mid-season 
plants, or were divided into current-year senescent tissue and the 
preformed bud for end-of-season plants. All aboveground tissues were dried 
at 70 degrees Celsius for 48 h.
The rhizome was weighed and split into three transverse sections of similar 
fresh weight (proximal, medial, and distal, or alternatively, from youngest 
to oldest). Each section was then split in half lengthwise. One half of 
each section was weighed and dried at 70 degrees Celsius for water content 
and total Kjeldahl N determination, while the remaining half of each 
section was stored at -20 degrees Celsius and used for later analysis.
Frozen tissues were ground in liquid N with a mortar and pestle. Samples 
(100-200 mg) were extracted with 1 ml 6% perchloric acid as described by 
Carpenter and Hand (1986). The insoluble pellet was dried at 70 degrees 
Celsius. Total amino acid concentration in the extracts was quantified 
using a ninhydrin method (Rosen 1957), measuring absorbance at 562 nm on an 
automatic plate reader (Molecular Devices Corporation). Analysis of 
individual amino acids was performed by high-pressure liquid 
chromatography (HPLC) on a C-18 column after derivitization of the samples 
with o-Phthalodialdehyde (Jones et al. 1982). Total Kjeldahl N 
concentrations of all tissues and fractions were determined as described by 
Jaeger and Monson (1992).
Identification of the non-protein amino acid was performed by H-NMR 
(solvent=D2O; frequency=300 MHZ; internal standard=TMS) and verified by 
acid hydrolysis of the amino acid and subsequent analysis by HPLC (as 
described above). The compound was extracted from frozen rhizomes ground in 
liquid N2 in 80% MeOH, and isolated using paper chromatography with a 
solvent system of phenol:H2O (4:1 w/w). Crystals were rinsed with diethyl 
ether. Acid hydrolysis was performed in 6 M HCl, at 100 degrees Celsisu for 
24 hr.
Averages for the entire rhizome (as determined from the proximal, medial,
and distal sections) are reported as "rhizome".
Fertilization treatment codes are 0 (control) and 1 (fertilized).
For date 090394, the "inflorescence" data are actually for the preformed 
bud. On date 091693, the preformed bud was included with the "shoot" data.
The 5 samples from the control plots for which delta-acetyl-ornithine
concentrations are reported, and actually collected on 060194, are
here reported as being collected on 052594 in order to allow comparison
between fertilized and control specimens.
The absence of some data for a given sample can indicate that the 
corresponding laboratory analyses were not performed, in addition to
being representative of "missing" data.
